Fisetin became of interest in a 2001 study screening for flavonoids able to prevent neuronal death induced by oxidative stress ; of about 30 compounds tested, only fisetin and quercetin were able to protect glutathione levels during stress. It was reborn in 2018 as a senolytic, that is, a chemical that kills senescent but not non-senescent cells. The former are not "dead cells" as claimed in this webpage at the time of this writing, but quite the opposite. They are not only alive but also resistant to die as expected, despite an accumulation of DNA damage and irreversible growth arrest. Cellular senescence is one key mechanism of aging and of age-related diseases. In mammals, the senolytic activity of fisetin has been verified in mice and in-vitro human tissues.[1] Source. The product is a smoke tree (Cotinus coggygria) extract powder 98% prepared by an undisclosed manufacturer ; extracts having 98% purity are widely available in the main Chinese wholesale online marketplace. The final product is sourced by the marketer Nature's Fusions (UT), and sold here under its Nutri label.[2] Clinical trials. Despite many marketing hyped claims about Mayo Clinic's fisetin clinical trials, their results are not published yet. At this time, a 2018 double-blind, randomized and placebo-controlled clinical trial in Iran (PubMed 29541713) seems the *only* published trial on fisetin as a senolythic ; there, colorectal cancer patients undergoing chemotherapy received 100 mg/day of either fisetin or placebo, and inflammatory markers IL-8 and hsCRP were found to be significantly reduced after seven weeks. Two of the Mayo's trials seem the most relevant to cellular senescence in older adults. These are pilot studies on the Alleviation of Frailty, Inflammation & Related Measures, one in women with gait issues, which started on 2/2018 (NCT03430037), and the other on inflammation markers in both women and men, which started on 11/2018 (NCT03675724). They were originally estimated to be completed in about 20 months, but their estimate dates have steadily been pushed forward every year over the last 5 years. At this time, their completion has mid 2024 estimates, and both are still listed as recruiting participants, despite that only 40 were needed for each study. Given the simple nature of these randomized trials, with very short time-frames for the primary outcome measure, and small sample sizes, something is clearly amiss.[3] Performance. This is the type of product for which a reviewer can only provide, at best, anecdotal and subjective before-and-after results. One person's results are not necessarily generalizable. Fisetin has a low aqueous solubility and poor oral absorption (a currently available liposomal format might improve the oral bioavailability, but that needs evidence-based studies), so supplementation outcomes with plenty of variability can be expected. YMMV.[4] Dosage. The dose of 20 mg/kg of body weight [~9 mg/lb] is indeed used in the Mayo Clinic's fisetin clinical trials, where it's given for 2 (or 3) consecutive days. However, the trials differ on what comes next -- two trials stop there ; two repeat the same dose a week later ; three repeat it a month later ; and one repeats it every four weeks over 20 weeks. The "hit-and-run" format of such an intermittent regime schedule is based on results obtained with different senolytics (dasatinib and quercetin) on genetically-modified mice, rather than fisetin on humans. As the outcome of these trials results have not been published yet, it is unclear if a hit-and-run dosage is as effective in humans as a daily supplementation with lower flavonoid amounts.[5] Risks. Fisetin clinical trials have not reported significant adverse effects so far, and no side effects with fisetin daily doses have been reported in the medical literature. Long-term use side effects in humans have yet to be determined.[6] Cost. My attached table compares the cost parameters of this brand to those of ten other brands of Fisetin extract with 500 mg or more per capsule, found on Amazon at the time of this review, which were anonymized by the letters A through J. I can't tell if it is doing me any good. I know that i did the research and felt i needed to add it to our vitamin intake. My choice. The seller sent it promptly.
